Barnsley were off the pace in the first half and Bradley Pritchard soon put Charlton ahead after an initial save by Luke Steele.
Johnnie Jackson drilled home the second after 19 minutes and Barnsley responded by bringing on Jason Scotland for Cywka.
Yann Kermorgant headed home soon after the break to leave the hosts facing à huge task.
Callum Harriott hit home the fourth before Stephen Dawson saw red for a doublé-footed lunge.
Sub Salim Kerkar made it five 10 minutes from time leaving victory against Derby on Tuesday even more important for the Reds.
To make matters worse, Tom Kennedy was also sent off for bringing down Ricardo Fuller.
Fuller added to Barnsley’s heaviest defeat of the season with the sixth in stoppage time.
